Abstract
In Mimulus verbenaceus, we identified three mutants with abnormal corolla development and significantly reduced nectar volume through EMS mutagenesis screening, designated as MV00081, MV00017, and NJ00356-1. BSA sequencing revealed that MV00081 corresponds to the homolog of the Arabidopsis STERILE APETALA. These mutants were named sap-1, sap-2, and sap-3, respectively. Among them, sap-1, caused by a premature termination mutation, exhibited the most pronounced phenotypic defects compared to the other two mutants. We measured leaf, flower, and nectar sugar content in sap-1 and found that it significantly inhibited leaf and petal development, reduced nectar production, and decreased the fructose and glucose content in nectar.
General Information
This dataset contains phenotypic measurements of different plant samples. The data (Supplemental_Table3.Phenotype_Measurements) is structured into six separate sheets, each representing a different aspect of plant morphology and physiology.
Dataset Description
1. Nectar Volume
- Description: This sheet records the nectar volume of different plant samples.
- Units: Microliters (μL).
- Columns: Each column represents a different plant sample, including MvBL, MV00081, MV00017, and NJ00356-1.
2. Leaf Measurements
- Description: This sheet provides measurements of leaf length and width.
- Units: Millimeters (mm).
- Columns: Each plant sample (e.g., MvBL, MV00081) has two associated columns: length and width.
3. Flower Width Measurements
- Description: This sheet contains data on the width of different flower parts.
- Units: Millimeters (mm).
- Columns:
- Dorsal Petal Width
- Lateral Petal Width
- Ventral Petal Width
- Corolla Tube Width
- Data is provided for multiple plant samples, including MvBL and MV00081.
- Data Organization:
- Each flower was measured for 17 biological replicates.
- Since each flower has two dorsal petals and two lateral petals, there are 34 data points for these measurements.
- Each flower has only one ventral petal and one corolla tube, resulting in 17 data points for these measurements.
- The lower number of data points for ventral petal width and corolla tube width is due to the biological structure of the flower, not missing data.
4. Flower Length Measurements
- Description: This sheet records the length of different floral structures.
- Units: Millimeters (mm).
- Columns:
- Calyx Length
- Dorsal Corolla Tube Length (DCT)
- Lateral Corolla Tube Length (LCT)
- Ventral Corolla Tube Length (VCT)
- Pistil Length
- Stamen Length
- Ovary Length
- Data is provided for multiple plant samples, including MvBL and MV00081.
5. Sugar Content
- Description: This sheet measures the concentration of sugars in different plant samples.
- Units: Milligrams per milliliter (mg/mL).
- Columns:
- Fructose
- Glucose
- Sucrose
- Data is available for samples such as MvBL and sap-1.
6. Length of 10 Contiguous Cells
- Description: This sheet records the length of 10 contiguous cells in different plant samples.
- Units: Micrometers (μm).
- Columns:
- Cell lengths measured for multiple samples, including MvBL and MV00081.
- Handling Missing Data:
- Missing data in the dataset is marked as "null".
- Some missing values are due to biological differences rather than data collection issues. For example, wild-type samples typically have 280 cells, whereas mutant samples have 220 cells. Missing values in these cases indicate absent cells in mutants rather than unrecorded data.
7. Flowering Time
- Description: This sheet records the number of days from sowing to floral bud emergence (defined as the appearance of a visible flower bud approximately 1 mm in size), as well as the number of leaves at that time point.
- Units: Days (for flowering time),Count (for leaf number).
- Columns:
- Days to floral bud emergence.
- Leaf number at floral bud emergence.
File Format
- The dataset is provided as an Excel file (.xlsx) containing six sheets.
- Each sheet has column headers indicating the measurement type and corresponding plant sample.

This Leaf.R script reads and processes leaf measurement data, summarizes it by treatment and index, and generates a bar plot with error bars and significance markers to compare leaf traits between two treatments.
This mv81_MvBL.R script analyzes floral organ length data from two treatments (MvBL and mv81), summarizes it by organ type, and generates a bar plot with error bars and significance markers to visually compare differences.
This mv81_MvBL_2.R script processes and summarizes petal width data from two treatments (MvBL and mv81), and generates a bar plot with error bars and significance markers to compare organ-level differences between treatments.
