Abstract
This dataset contains the source data supporting the article entitled Excitation-contraction coupling, cardiomyocyte electrophysiology, and transcriptome profiles in two HFpEF murine models: Etiology and sex-dependent differences. The dataset supports key findings in recent two-hit translational animal models of heart failure with preserved ejection fraction (HFpEF), including the hypertensive, nitrosative-stressed mice fed with high-fat diet and L-NAME (HFD+L-NAME) and the obese-diabetic leptin receptor-deficient db/db mice with excess aldosterone (db/db+Aldo), which may phenocopy select subgroups of HFpEF. Morphometry, echocardiography, electrophysiology (action potentials and ionic currents), fluorescent Ca imaging, and RNA sequencing data are included in separate Excel sheets with individual cell data and group statistics. The dataset is structured to follow the sequence of data presented in the Figures in the article and to ease the navigation between the uploaded data files.

Description of the data and file structure
This dataset contains morphometry, echocardiography, electrophysiology (action potentials and ionic currents), fluorescent Ca imaging, and RNA sequencing data in separate Excel sheets with individual cell data and group statistics. Files are compressed to keep the folder structure and to ease navigation between the uploaded documents. Description: This compressed file contains two Excel files with action potential (AP) data in 3 groups of experimental animals, including (1) control sham, (2) leptin receptor-deficient db/db mice with chronic aldosterone infusion (db/db+Aldo), and (3) high-fat diet combined with nitric oxide synthase inhibitor L-NAME treatment (HFD+L-NAME). Male and female data are reported separately in each group.
"Fig3_data_AP-parameters.xlsx" provides ventricular cardiomyocyte AP data at 1 Hz steady-state pacing frequency. Sheet "Panel A" provides typical AP waveforms data (time in milliseconds and membrane voltage in millivolts). Sheet "Panel B" provides AP repolarization parameters, including AP duration at 20, 50, and 90% of repolarization (APD20, APD50, and APD90, respectively) in milliseconds. Sheet "Panel C" provides additional AP parameters, including resting membrane potential (RMP, in millivolts), maximal rate of AP upstroke velocity (dV/dtmax, in volts per second), and maximal rate of AP phase 3 repolarization (-dV/dtmax, in volts per second).
"Fig4_data_Frequency-STV-Alternans.xlsx" provides ventricular cardiomyocyte AP data at various pacing rates from 1 Hz to 10 Hz. Sheet "Panel A" provides data about the frequency dependence of AP duration at 90% of repolarization (APD90, in milliseconds). Sheet "Panel B" provides typical AP waveforms data at a 10 Hz pacing rate (time in milliseconds and membrane voltage in millivolts). Sheet "Panel C" provides fifty consecutive APD90 data at a 1 Hz pacing rate (time in seconds, APD90 in milliseconds). Sheet "Panel D" provides the short-term variability of APD90 at 1 Hz pacing (STV, in milliseconds), the magnitude of APD90 alternans (difference between long and short beats) at 10 Hz pacing (in milliseconds).

Male and female data are reported separately in each group.
"Fig10_data_CaT.xlsx" provides ventricular cardiomyocyte intracellular Ca2+ transient (CaT) data. Sheet "Panel B" provides typical caffeine-induced Ca2+ transient (Caff T) data (time in milliseconds and fluorescence expressed as background corrected F/F0, where F0 is the non-paced baseline fluorescence). Sheet "Panel C" provides CaT parameters data at 1 Hz steady-state pacing, including CaT amplitude (difference between baseline and peak fluorescence, normalized to non-paced F0), CaT tau decay (in milliseconds), and sarcoplasmic reticulum (SR) Ca2+ load assessed by rapid caffeine application (difference between baseline and peak fluorescence, normalized to non-paced F0). Sheet "Panel D" provides CaT parameters at 2 Hz steady-state pacing, including CaT amplitude (difference between baseline and peak fluorescence, normalized to non-paced F0), CaT tau decay (in milliseconds), and Caff T decay slope (change in fluorescence per second).
"Fig11_data_Ca-sparks-and-waves.xlsx" provides diastolic Ca2+ spark data in ventricular cardiomyocytes. Sheet "Panel B" provides Ca2+ spark frequency (number of Ca2+ sparks per 100 micrometers per second) and Ca2+ wave frequency (number of waves per minute).

Sheet "Panel C" provides the density of IK1 (IK1 magnitude normalized to cell capacitance) at -140 millivolts and at -40 millivolts (in picoampere per picofarad).
"Fig7_data_IKv.xlsx" provides data on the voltage-gated potassium currents (IKv). Sheet "Panel A" provides typical IKv traces (time in milliseconds and IKv current in picoampere per picofarad).
