Abstract
This work describes the development and validation of a high-sensitivity analytical method (LC-MS/MS IAM-TPP) for the simultaneous quantification of protein persulfides, low molecular weight persulfides/polysulfides and hydrogen sulfide (H₂S) in complex biological matrices. The method was applied to biological samples to characterise H2S and endogenous persulfide/polysulfide formation, including analysis or physiologically relevant ischemia reperfusion models. The raw dataset is intended to enable independent re-analysis, method reuse, and further translational development in redox biology and drug discovery.

No open-source or publicly available software was used for data generation or analysis.
The raw LC–MS/MS data files can be opened and processed only using the Waters MassLynx software and associated QuanLynx processing module, which are proprietary to Waters Corporation.
No additional scripts and/or custom code were used or are available beyond the manufacturer’s software environment.

All data associated with this study are original and generated by the authors. No data were obtained or derived from external sources. There are no additional publicly accessible datasets beyond those deposited in this Dryad repository and the supporting information available through the Nature Communications publication.
The data derive from experiments described in the manuscript: using the IAM-TPP technique, persulfide and polysulfide species were trapped, derivatised, separated by liquid chromatography, and quantified by tandem mass spectrometry. Sample preparation, calibration curves, and validation experiments (linearity, limit-of-detection, reproducibility) are detailed in the publication.
Limitations: The dataset reflects specific sample types, instruments, and experimental conditions; normalisation and instrument-specific settings may differ in other laboratories. Raw data are provided to enable inspection of processing pipelines.
