Abstract
As the early step of food ingestion, the swallow is under rigorous sensorimotor control. Nevertheless, the mechanisms underlying swallow control at a molecular and circuitry level remain largely unknown. Here, we find that mutation of the mechanotransduction channel genes nompC, Tmc, or piezo impairs the regular pumping rhythm of the cibarium during feeding of the fruit fly Drosophila melanogaster. A group of multi-dendritic mechanosensory neurons, which co-express the three channels, wrap the cibarium and are crucial for coordinating the filling and emptying of the cibarium. Inhibition of them causes difficulty in food emptying in the cibarium, while their activation leads to difficulty in cibarium filling. Synaptic and functional connections are detected between the pharyngeal mechanosensory neurons and the motor circuit that controls swallow. This study elucidates the role of mechanosensation in swallow, and provides insights for a better understanding of the neural basis of food swallow.

Description of the data and file structure
This dataset includes raw data of confocal microscopy and calcium imaging, which was used in the figures of https://doi.org/10.7554/eLife.88614
Analysis file was built in GraphPad 8, which is associated with behavior experiment and calcium imaging.
Raw data of behavior videos (60fps avi video of Drosophila behavior) are 386GB, and we offer access to them in the behavior file description.

Tmc-GAL4, nompC-QF>>mCherrry/GFP/RedStinger; among them, cibairum muscle of fly was dyed with phalloidin in file named tmc(ii)-nompc-flp-out-GFP-cibarium-1 (figure4)
Tmc, nompC, piezo, Tmc&piezo, nompC&piezo > GFP;
Tmc-GAL4, nompC-QF>>GFP; and labellum of fly was ablated.

File naming convention: Folder name inside indicates which figure the data was used in. File name inside each second folder indicates the genotype of Drosophila, and its body part (brain or cibarium/VCSO, sometimes was abbreviated to B or C/v). Tmc-GAL4,nompC-QF driver was abbreviated to TN. The number in the end of file name indicates the order number of candidate sample in the day's microscopy.

Description: The folder consists of raw data for calcium imaging used in figure4D-K of the article. It is about calcium imaging in motor neuron 12/11 while activating md-C neurons , and calcium imaging in md-C neurons while fly is swallowing. It includes:
Tmc-GAL4>P2X2, MN11/12>GCaMP6s; and control
Tmc-GAL4, nompC-QF>>GCaMP6m while swallowing; and control
File naming convention: "MN11" folder includes raw data of Figure4G-I, "MN12" folder includes raw data of Figure4D-F, they are about calcium imaging in motor neuron 12/11 while activating md-C neurons, and folder name inside indicates the genotype and added stimuli (water or ATP). "TN-gcamp6" folder includes raw data of Figure4J-K, where fly's genotype is Tmc-GAL4,nompC-QF (abbreviated to TN)>>GCaMP6m.
