Tab "result summary" This tab summarizes the results for all Etv4-MADM11 clones and for all control MADM11 clones (the numbers are calculated on the following tabs). The numbers in cells I6:K6 are plotted in Fig 4H and listed as "Combined data" in the Table in Fig 4I. The numbers in cells I2:K4 are also listed in the Table in Fig 4I. The numbers in cells I12:K12 are plotted in Fig 4J. Column A lists the embryo genotypes for Etv4 and Etv5. Columns B and C list the genotypes of green and red cells. Column D lists the number of clones in which Etv4+/+ cells were closer to the tips at the end of the time lapse sequence (for Etv4-MADM11 clones) or in which green cells were closer to the tips (for control clones). Column E lists the number of clones in which Etv4-/- cells were closer to the tips at the end of the time lapse sequence (for Etv4-MADM11 clones) or in which red cells were closer to the tips (for control clones). Column F lists the number of clones in which neither color/genotype of cells was clearly closer to the tips. Column G lists the total number of clones of each genotype. Column H is blank. Columns I-K list the percentages of clones in which green cells, red cells, or neither were closer to the tips at the end of the time lapse sequence. Tab "all Etv4-MADM11 kidneys". This tab lists the data for all Etv4-MADM11 kidneys (including all three possible Etv5 genotypes) in decreasing order of total ending cell number. Column A lists the clone identity. Column B lists the Etv5 genotype. Columns C-E indicate whether WT, mutant (Etv4-/-) or neither cell type was closer to the tip at the end of the time-lapse sequence. Totals are listed at the bottom. Column F is blank. Columns G-J list the starting and ending number of WT or mutant (Etv4-/-) cells in each clone analyzed. Average and SD are calculated at the bottom. For clones labeled "can't count" the final cell number could not be determined for one of several reasons (e.g., poor focus at end, part of clone moved out of frame, etc.). Column K lists the ending number of total cells in each clone analyzed. Columns L and M list cell cycle lengths for the WT and mutant cells, when they could be measured, and these are tabulated in the tab "cell cycle lengths". The next three Tabs list the same data for the Etv4-MADM11 clones, segregated by the Etv5 genotype (Etv5+/+, Etv5+/M or Etv5M/M). Tab "Etv4+- Etv5++ kidneys" Columns A-K are the same as defined for Tab "all Etv4-MADM11 kidneys" Tab "Etv4+- Etv5+M kidneys" Columns A-K are the same as defined for Tab "all Etv4-MADM11 kidneys" Tab "Etv4+- Etv5MM kidneys" Columns A-K are the same as defined for Tab "all Etv4-MADM11 kidneys" Tab "cell cycle length" Column A is blank Columns B and C list the individual cell cycle times measured in Etv4+/-, Etv5+/+ embryos for green (Etv4+/+) or red (Etv4-/-) cells. Average and SD are calculated at the bottom. Column D is blank. Columns E and F list the individual cell cycle times measured in Etv4+/-, Etv5+/M embryos for green (Etv4+/+) or red (Etv4-/-) cells. Average and SD are calculated at the bottom. (For the Etv4+/-, Etv5M/M kidneys, there were too few cell cycle times that could be measured to generate meaningful statistics.) Column G is blank. Columns H and I list the combined data from columns B, C, E and F, and calculate the combined average and SD at the bottom. Tab "control MADM11 kidneys" This lists the data for control MADM11 clones, from kidneys which were either Etv4+/+,Etv5+/+ (37 clones) or Etv4+/+,Etv5+/M (4 clones). Column A lists the clone identity. Column B lists the Etv5 genotype. Columns C-E indicate whether green, red or neither cell color was closer to the tip at the end of the time-lapse sequence. Totals are listed at the bottom. Column F is blank. Columns G-J list the starting and ending number green WT or red cells in each clone analyzed. Average and SD are calculated at the bottom. For clones labeled "can't count" the final cell number could not be determined for one of several reasons (e.g., poor focus at end, part of clone moved out of frame, etc.). Column K lists the ending number of total cells in each clone analyzed. For questions contact Frank Costantini fdc3@columbia.edu
