Abstract

This data set is designed for cell contour and cell track analysis. Hence image sequences of moving Dictyostelium discoideum cells are recorded. For the purpose to facilitate the detection of the cell contour we take fluorescent images of the cortical protein actin to obtain a high contrast between background and cell body. In each image sequences several cells are recorded. This allows for an analysis of all cells at once or to crop single cell tracks.

Methods

Fluorescent images sequences are recorded with a confocal laser scanning microscop. The sequences have the length of 500 and 1000 frames. The time interval is one second. All images have a resolution of  512 x 512 pixel and are 8 bit. The physical resolution is 134.95 x 134.95 µm and  212.55 x 212.55 µm. In preparation of the experiments the cells are staved for 4.5 hours in buffer to induce chemotaxis.
Actin polymerization is visualized by using actin binding LifeAct-mRFP.

Usage notes

The data format is accessible with the free software ImageJ (Fiji) and with Zeis Zen software (there is a limited free version available).
