README Senescence screen of the Kronos TILLING population Referred to in Harrington et al. 2019 doi: 10.3389/fpls.2019.00963 The associated Excel file contains all phenotypic scores for heading, flag leaf senescence, and peduncle senescence of the Kronos TILLING population. The trial was carried out in 2015 at the John Innes Centre in Norwich, UK. Relevant information on trial design and field management practices are available in the associated paper (Harrington et al. 2019) Two tabs are contained in the excel file: 1- "Senescence Scoring" This tab includes the name of each TILLING line phenotyped ("Kronos TILLING Line"), as well as the relative position in the field trial ("Individual plot label"). All TILLING lines were phenotyped for heading date (scored upon 75% of spike emergence); Flag leaf senescence date (scored when 25% of the flag leaf showed senescence); and Peduncle senescence date (scored when the top 1 inch/3 cm of the peduncle had turned yellow). We also noted whether a senescence phenotype appeared to segregate within the plot, suggesting that the relevant mutation may be heterozygous in the TILLING line ("Is a senescence phenotype segregating in the plot?"). Those which did appear to be segregating were marked "seg", as were some plots which may or may not have shown segregationg ("maybe/unclear") Finally, the number of days elapsed between heading and both flag leaf and peduncle senescence for each TILLING line was also calculated ("Days from heading to flag leaf/peduncle senescence"). 2- "Field Plan" This tab shows the field plan used for the TILLING trial. All lines were sown as unreplicated single 1 m rows in 6 x 1 m plots (i.e. each plot contains 6 independent 1 m rows, and thus 6 independent TILLING lines). All racks contain 20 plots, with the exception of rack 1 which includes an additional two hand-dibbed plots between plots 17 and 18. Within the field plan, the Kronos TILLING ID number is given for each row, allowing comparison of senescence timings with relative plot location if desired.
