The data include results from Enzyme-Linked Immunosorbent Assay (ELISA) of NADPH oxidases (NOX1, NOX2 and NOX4) and asymmetric dimethylarginine (ADMA) of healthy and hypertensive patients (details are included in Elbatreek et. al., 2020). After the article "NOX5-induced uncoupling of endothelial NO synthase is a causal mechanism and theragnostic target of an age-related hypertension endotype" was published, concerns were raised about repeated values in the datasets underlying Figures S1A. We provide this file to explain the reasons for the repeated values. The values provided for S1 Fig. in the article’s S1 Data file are ng/mL values calculated from optical density (O.D.) measurements. The dataset contains repeated values for S1A Fig. because NOX1 concentrations were very close to the limit of detection, i.e., close to the O.D. of the buffer control for which we obtained a value of 0.09. The repeated values in the 0.07-0.16 range were interpreted as essentially negative results, and from these data we concluded that NOX1 was not detectable using this assay for most subjects. Only one normotensive and two hypertensive patients with microalbuminuria yielded O.D. readings above the 0.07-0.16 range. 2. Relationship between files, if important: N/A 3. Are there multiple versions of the dataset? no METHODOLOGICAL INFORMATION All procedures were approved by the research ethics committee of Taipei Veterans General Hospital (Taipei Veterans General Hospital Institutional Review Board, No.: 96-12-42A), and all participants provided their written informed consent. The study complied with the Declaration of Helsinki. It was approved by the appropriate health authorities, an independent ethics committees, and the Institutional Review Board of Taipei Veterans General Hospital. Venous blood samples were collected from all patients after 8 hours of overnight fasting for measurement. The blood samples were centrifuged at 3,000 rpm for 10 minutes immediately after collection, and the plasma samples were kept frozen at −70°C until analysis. Each standard and plasma sample was analyzed twice, and the mean value was used in all subsequent analyses. Plasma ADMA levels were measured by ADMA Fast Enzyme-Linked Immunosorbent Assay (ELISA) kit (DLD Diagnostika GMBH, Hamburg, Germany). CD144+ microparticles were isolated as described with modifications (Elbatreek et. al., 2020). Briefly, Dynabeads G (Invitrogen, Carlsbad, CA) were washed with PBS containing 0.1% BSA and then reconstituted with PBS. Anti-CD144 antibody (Santa Cruz Biotechnology, Dallas, TX), which specifically targets endothelial cells, was mixed with prewashed Dynabeads G for 2 hours and then incubated with plasma samples at 1:200 dilution overnight at 4°C. After precipitation, Dynabeads G were washed with PBS and 1% Tween-20 three times. The purity of CD144+ MPs, determined by FACS analysis, was 70% ± 5.6%. With the use of FITC-conjugated beads as size references, the size of such particles was assessed to be <0.5 μm in diameter. Human NOX1, NOX2 and NOX4 levels were measured using commercially available ELISA (Cloud-Clone, Katy, TX) kits according to the manufacturer’s instructions.
