Nature Communications. GENERAL NOTES: Three data files are associated with this paper. Each file contains the data associated with one experiment. This ReadMe file pertains to all three data files associated with this paper. EXPERIMENT 1 DATA FILENAME: Alonzo_et_al_Nature_Comm_2016_Expt1.csv This file contains the data from Experiment 1 (the ?in vitro? sperm competition experiments. These data are represented in Figures 1 and 2 and Tables 1 and 2. Each column represents a different variable (described below) Each row represents a different experimental replicate (i.e. in vitro sperm competition trial or cross) Variable Description (i.e. Column heading in the file followed by a description of the variable): In_Vitro_Cross_Number: This name designates the experimental cross for bookkeeping and sample identification. Ovarian_Fluid_Treatment: treatment- either Rinsed, Ovarian or Baseline, see text for details) Number_Gentoyped: The number of offspring genotyped Number_Assigned_NM: The number of offspring assigned to the nesting male in the genetic paternity analyses SN_sperm_number: The average number of sperm found in a subsample of the sneaker male?s sperm sample (for that in vitro cross number). NM_sperm_number: The average number of sperm found in a subsample of the nesting male?s sperm sample (for that in vitro cross number). The following prefixes represent the sperm characteristics for sneaker males 40 seconds after activation (SN1-) nesting males 40 seconds after activation (NM1-) sneaker males 50 seconds after activation (SN2-) nesting males 50 seconds after activation (NM2-) sneaker males 60 seconds after activation (SN3-) nesting males 60 seconds after activation (NM3-) The sperm characteristics (as identified by Hamilton Thorne?s Animal Motility CASA software) for each sample are given by the following variable designations: VAP (average path velocity of sperm cells) VSL: (straight line velocity of sperm cells) VCL: (curvilinear velocity of sperm cells) STR: (straightness of sperm cells=VSL/VAP) LIN: (linearity of sperm cells=VSL/VCL) TOTAL_COUNT: (total count of sperm cells) MOTILE_COUNT: (total count of motile sperm cells) EXPERIMENT 2 DATA FILENAME: Alonzo_et_al_Nature_Comm_2016_Expt1.csv This file contains the data from Experiment 2 (the experiment examining the effect of ovarian fluid on sperm characteristics). These data are represented in Figure 3. Each row in the data file represents a new replicate in the experiment (i.e. sperm form an individual male) Variable Description (i.e. Column heading in the file followed by a description of the variable): NAME: This name designates the experimental replicate for bookkeeping and sample identification. MALETYPE: Designates whether the sperm sampled was collected from either a sneaker male (SN) or a nesting male (NM) OF/SW : Treatment type OF=with ovarian fluid, SW=seawater without ovarian fluid VAP: as for experiment 1 above VSL: as for experiment 1 above VCL: as for experiment 1 above STR: as for experiment 1 above LIN: as for experiment 1 above TOTAL_COUNT: as for experiment 1 above MOTILE_COUNT: as for experiment 1 above REPEAT: Number of minutes since activation of the sperm sample. MOTILITY: The proportion of sperm cells as identified by the CASA program that are motile or zero if no sperm cells were motile (motile count/total count). EXPERIMENT 3 DATA FILENAME: Alonzo_et_al_Nature_Comm_2016_Expt1.csv This file contains the data from Experiment 3 (the experiment examining the effect of ovarian fluid on sperm chemoattraction). These data are represented in Figure 4. Each row in the data file represents a new replicate in the experiment (i.e. sperm form an individual male) Variable Description (i.e. Column heading in the file followed by a description of the variable): Assay_Number: This name designates the experimental replicate for bookkeeping and sample identification. Male_Type: Designates whether the sperm sampled was collected from either a sneaker male (SN) or a nesting male (NM) Treatment: Experimental treatment = EGGS (ovarian fluid and eggs in seawater in the bottom chamber) or SW (seawater only in the bottom chamber) Sperm_Number_Top: Number of total sperm cells estimated to be in the top sperm samples (i.e. above the Transwell membrane) Sperm_Number_Bottom: Number of total sperm cells estimated to be in the bottom sperm samples (i.e. below the Transwell membrane, i.e. the cells that crossed the membrane)
