Supplementary file 1. How to upload P. falciparum Ribosome Profiling MochiView tracks. 1. Download Mochiview http://johnsonlab.ucsf.edu/mochiView/software-tutorial-download.html Software requirements: Java 7 2. Open Mochiview v1.46 3. Download the database containing all the ribosome profiling data files (wig files): ribosomeprofilingMOCHIVIEW.cvw 4. Load the database: Click on the import tab -> Database -> Manage Database 5. From the Manage Databases pop up screen, press Import and click on the ribosomeprofilingMOCHIVIEW.cvw file downloaded in part 3 & press open. This step will take a few minutes to upload. Hit the green check button once the upload is successful. 6. Press the Database tab and select the Malaria Ribosome Profiling Database. Click Activate to unzip the files. Restart Mochiview and the database will be fully loaded. Data tracks: (all are scaled to 0-100 or -100 to 0) From the top to bottom: Ribosome footprint read coverage plots (normalized by millions of reads sequenced) mRNA read coverage plots (normalized by millions of reads sequenced) Timepoints are referred to as 1-5 (Rings, Early Trophs, Late Trophs, Schizont, Merozoites, respectively) Gene Tracks 30mer mappability track Navigating the datafiles: To switch chromosomes: press ALT + L and select the chromosome from Select Sequence drop down menu To scroll through a chromosome: drag the bar on the bottom right of the screen
