Abstract
Tauopathies, such as Alzheimer’s disease and Frontotemporal Dementia, are common neurodegenerative diseases characterized by misfolding, hyperphosphorylation, and aggregation of Tau. Molecular mechanisms underlying Tauopathies are still poorly understood, in part due to a lack of human models endogenously developing major disease hallmarks. Adult Tau isoform expression contributes to Tau pathogenesis but is challenging to replicate in human stem-cell-derived systems, which impedes the formation of late-stage disease phenotypes and, hence, the research on underlying mechanisms and drug development. Here, we show that induction of adult human brain-like 4R Tau isoform expression enables endogenous formation of late-stage Tauopathy hallmarks in iPSC-derived neurons engineered to contain synergistic Tau mutations. Neurons accumulated seeding-competent, hyper-phosphorylated, fibrillar Tau in tangle-like structures. Furthermore, exclusive expression of mutant 4R in the absence of the 3R Tau isoform disproportionately intensified pathology, resulting in highly abundant Tau misfolding and aggregation. Finally, we provide proof-of-principle that our model can be translationally applied both to test chemical disease modulators and evaluate a human Tau PET tracer. Collectively, our model enables novel investigations on endogenous mechanisms of human Tauopathy formation, suggesting a central role of 4R Tau isoform expression for pathogenesis in human neurons. Moreover, it may also serve as a platform supporting urgently needed development of disease-modifying drugs.

Description of the data and file structure
Data files contain sequencing traces for off-target quality control of multiple cell lines created in the linked study by CRISPR-editing. Sequencing traces of the top predicted off-target loci for each gRNA used for editing in this study are included. Included are the most likely off-target binding sites for each gRNA predicted by MIT and CFD algorithms in CRISPOR (https://crispor.gi.ucsc.edu) in both .ab1 and .fasta format.
As a reference, each predicted off-target site was sequenced in WT cells before CRISPR editing, and the sequencing traces are included to allow for easy comparison.
Code/software
The .ab1 files contain the sequencing result,s including intensities, and need to be opened using a sequencing data analysis software (such as SnapGene, VectorNTI, or CutePeaks (free and open-source)). Off-target effects in these sequencing traces would be characterised by a clean sequence trace (before the predicted cut site) suddenly changing into a double peak pattern (around and after the cut site).
We also provide .fasta files, which are plain text files and can be opened using any simple text editor to ensure no special software is needed. They only contain the sequence itself without information on heterozygosity, etc. They can be easily used to confirm that the traces overall match the WT sequence (demonstrating the absence of homozygous indels) and to exclude the presence of large stretches of bases that could not unequivocally be identified (such as S, K, M, R, Y, etc), which would be indicative of heterozygous off-target effects.
