Abstract
Most insects, including agricultural pests and disease vectors, rely on olfaction for key innate behaviors. Consequently, there is growing interest in studying insect olfaction to gain insights into odor-driven behavior and to support efforts in vector control. Calcium imaging using GCaMP fluorescence is widely used to identify olfactory receptor neurons (ORNs) responsive to ethologically relevant odors. However, accurate interpretation of GCaMP signals in the antenna requires understanding both response uniformity within an ORN population and how calcium signals relate to spike activity. To address this, we optimized a dual-modality recording method combining single-sensillum electrophysiology and widefield imaging for Drosophila ORNs. Calcium imaging showed that homotypic ab2A neurons exhibit similar odor sensitivity, consistent with spike recordings, indicating that a single ORN’s response can reliably represent its homotypic counterparts. Furthermore, concurrent dual recordings revealed that peak calcium responses are linearly correlated with spike activity, regardless of imaging site (soma or dendrites), GCaMP variant, odorant, or fly age. These findings validate the use of somatic calcium signals as a reliable proxy for spike activity in fly ORNs and provide a foundation for future large-scale surveys of spike–calcium response relationships across diverse ORN types.

Description: This Excel file contains non-formatted source data for Figure 3. All formatting was removed and empty cells are replaced with NaN to improve accessibility. The workbook includes paired electrophysiological and calcium-imaging responses from Drosophila ab2A/Or59b olfactory receptor neurons expressing different genetically encoded calcium indicators (GCaMP6m, GCaMP7c, and GCaMP8s). Responses were measured for two odorants, methyl acetate and ethyl acetate, across a range of stimulus concentrations.
Variables

Left table (methyl acetate):
- Peak spike response (Hz): Maximum odor-evoked action potential firing rate measured by single-sensillum recording (SSR), in spikes per second.
- methyl acetate dilution: Odor stimulus concentration represented as decimal dilution fractions (e.g., 0.000001 = 10⁻⁶ dilution).
- Fly1–Fly7: Independent biological replicates.
- AVE: Mean response across biological replicates.
- SEM: Standard error of the mean across biological replicates.
Right table (ethyl acetate):
- Peak spike response (Hz): Maximum odor-evoked action potential firing rate measured by SSR.

Variables
For each GCaMP variant (GCaMP6m, GCaMP7c, GCaMP8s):
- R2: Coefficient of determination for the linear regression between spike response and calcium fluorescence response.
- Linear relationship: Best-fit regression equation, where x is spike response (Hz) and y is calcium fluorescence response (ΔF/F, %).
- P value: Statistical significance of the regression.
- One-way MANOVA statistics:
- Comparison: Multivariate comparison between methyl acetate and ethyl acetate response profiles for each GCaMP variant.
- P value: Statistical significance of the comparison.
