Mitochondrial reference genome for Teleopsis dalmanni

Abstract

We assembled a complete mitochondrial genome using publicly available PacBio HiFi reads, which were generated from pooled Teleopsis dalmanni larvae. The assembly was conducted using MitoHiFi v3.01 (Uliano-Silva et al., 2023), a tool designed for accurate mitochondrial genome assembly from high-fidelity long-read data. Specifically, we used the raw HiFi reads as input and employed the mitochondrial genome of the rust fly (Loxocera sinicia) as a reference to guide the assembly. Genome annotation was carried out using MitoFinder, which identified the full complement of mitochondrial genes. The final result was a circularised mitochondrial genome assembly measuring 20,708 base pairs in length and containing all 37 typical mitochondrial genes, including 13 protein-coding genes, 22 tRNAs, and 2 rRNAs.
