Abstract
This dataset presents electrophysiological properties from whole-cell patch clamped nucleus accumbens core medium spiny neurons from male rats and female rats recorded in different estrous cycle phases. Excitatory synaptic input, action potential, and intrinsic properties are presented in an analyzed format, as well as the original electrophysiological recordings of each neuron. These data were originally presented across two different papers (Proaño et al., 2018, 2020) and further analyzed in an additional paper (Long et al., 2024), all of which are cited below. These data were key for establishing that nucleus accumbens neuron electrical function changed across the estrous cycle in response to its associated hormones, programming a phase-dependent sex difference in adulthood. This finding provided a new framework for understanding how biological sex and hormone cyclicity regulate the neurons implicated in motivated behaviors and related functions and disorders.

The excel spreadsheet features whole-cell patch clamp electrophysiology data of nucleus accumbens core medium spiny neuron properties of adult male and female rats. Data were collected in males and in females of different estrous cycle phases in females. Some pieces of data are not available and are indicated in the spreadsheet with ‘N/A’. For ‘N/A’ values in mEPSC of amplitude, frequency, and decay, values are not present because these neurons were not recorded in this analysis. For values not available in delay to first action potential, greater than two spikes were recorded and thus not included in the study. Please see the cited papers for methodological details.
There are two types of files associated with this repository. The excel spreadsheet contains the analyzed electrophysiological characteristics of all neurons. The name of the excel spreadsheet is: Neuron data. The second type of file contains the raw data obtained from the whole-cell patch clamp recordings of individual neurons. These files are generated from PClamp and have an axon binary format (.abf) extension. The files can be read by PClamp, Clampfit, or can be accessed through the pyABF, ABFview, AbfConvert, abftools, python or R packages/toolkits (https://github.com/topics/abf). The ABF files associated with individual neurons are named starting at Excel file sheet 3.
File Overview:
A) Neuron data.xlsx
This file is an excel spreadsheet containing analyzed neuron electrophysiological data. Sheets within the file:
1) Excel file sheet 1 – All Cells: Summary of all collected data for electrophysiological properties of medium spiny neurons obtained from adult male and female rat nucleus accumbens core.
2) Excel file sheet 2 – Analysis Template: Blank template used to analyze data collected from individual medium spiny neurons.
3) Excel file sheets 3-End - Starting at Excel file sheet 3 are sheets dedicated to the analysis of recordings from each individual Medium Spiny Neuron for this project. Each tab is named by the date (MMDDYY) plus N and a number indicating the number of the cell recorded on that date. For example, 040419N1 would be the first cell recorded on 04/04/19, whereas 040419N2 would be the second cell recorded on 04/04/19.
B) .abf Files:
There are 1483 .abf files associated with this project. Data are organized into two zipped folders, one containing mEPSC data and the other containing additional electrophysiological properties. Within the zipped folder, individual folders are named by the date of recording: take folder 040419 as an example (Cells recorded on 04/04/19). Within each date folder, is a folder dedicated to an individual neuron recorded on that date, in the 040419 folder are 040419N1, 040419N2, …, 040419N6 (Each folder contains the files for the respective recording). The folders for each individual neuron are named as stated on the “Neuron data” excel spreadsheet. Within each of these folders are the .abf files associated with the recordings of that individual neuron. The identity of the .abf files used for analyzing specific neuron electrophysiological properties are defined on the respective excel sheet.
Version Changes:
11/22/24: Added abf files “neuron_electrophysiology_mEPSC_abf_files.zip” which contain the mEPSC data. Updated Excel file sheet to include names of files containing mEPSC data for each respective recorded neuron.
This dataset was collected using whole-cell patch clamp recordings of medium spiny neurons in adult male and female rat nucleus accumbens core. The recordings were analyzed to generate standard attributes describing the biophysical properties of these neurons and were compared by sex and female estrous cycle phase.
