Abstract
Male genitalia are usually extremely divergent between closely related species, but relatively constant within one species. Here, we examine the effect of temperature on the shape of the ventral branches, a male genital structure involved in reproductive isolation, in the sister species Drosophila santomea and D. yakuba. We designed a semi-automatic measurement machine learning pipeline that can reliably identify curvatures and landmarks based on manually digitized contours of the ventral branches. With this method, we observed that temperature does not affect ventral branches in D. yakuba, but that in D. santomea ventral branches tend to morph into a D. yakuba-like shape at lower temperatures. We found that male genitalia structures involved in reproductive isolation can be relatively variable within one species and can resemble the shape of closely related species’ genitalia through plasticity to temperature. Our results suggest that reproductive isolation mechanisms may be dependent on the environmental context.

contours.zip
The contour folder contains all contour measures obtained via the custom ImageJ pluggin. Each subfolder is one isofemale line at one specific temperature following the nomenclature of Table 1 (list of isofemale lines) and each .txt file is a tab separated file where each line is the x (first column) and y (second column) coordinate of the pixel under the contour.
pictures.zip
The picture folder contains the corresponding pictures for the contour files using the same nomenclature.
parametrization.zip
This folder contains the pictures, contour and hand-positionned landmarks (.tps file) for D. santomea strain 1563 and D. yakuba Raphia. For D. yakuba Raphia, other contours can be found in the countour folder.

This java script is a plugin to ImageJ in order to record the coordinates of each pixel along the drawn contour on ImageJ. This plugin requests an input directory that contains the pictures to be analyzed or sub-directories containing pictures that will be successively analyzed. The output is a tab-separated file, for each picture (here individuals), with the x (first column) and y (second column) coordinates of the points where the contour passes.
Wait_For_User.java
This java script plugs to the contour java script and triggers a ask-user window before opening the next picture in the stack.
