Abstract
Our study investigates transgenerational immune priming in three clones of Daphnia dentifera. We collected offspring from mothers either unexposed or exposed to pathogens and then exposed offspring to either the same pathogen as the maternal exposure or a different pathogen. We measured offspring susceptibility for all treatments and pathogen burden for the two ‘obligate killer’ pathogens. We did not find any evidence of transgenerational immune priming. This suggests that transgenerational immune priming is not a strong factor influencing disease dynamics in the D. dentifera system.

Description of the data and file structure
Data and code for a study investigating transgenerational immune priming in three clones (genotypes) of Daphnia dentifera. The three clones are named Standard ('S'), BD08-46 ('B'), and DW22-58 ('D'). They are known to be susceptible to the three pathogens used in this study, although the degree of susceptibility differs across clones. We measured offspring susceptibility to three different pathogens and offspring pathogen burden.
Files and variables
File: B_Clone.csv
Description: This data file contains the infection status and spore burden data for the B clone.
Variables
- Replicate: replicate number of individual
- GeneralMaternalExposure: general maternal pathogen exposure treatment (none = mother was not exposed to any pathogens; pathogen = mother was exposed to a pathogen).
- MaternalParasite: specific maternal pathogen treatment, specifying the pathogen species when applicable
- CurrentParasite: specific pathogen exposure treatment of offspring generation. BirthDate = date the individual was born
- BirthDate: date the individual was born
- DeathDate: date the individual died (NA means that the individual was terminated at the end of the experiment to determine infection status)
- InfectionStatus: infection status of the individual (0 = uninfected, 1 = infected, NA = unable to determine infection status due to early host death or handling error)
- Burden1: first mature spore count of spore slurry from individual, conducted with hemocytometer
- Burden2: second mature spore count of spore slurry from individual, conducted with hemocytometer
- AvgMatureBurden: average mature burden of individual, calculated from Burden1 and Burden2
- SporesPerHost: calculation of mature spores per individual based on the individual being preserved in 100 microliters of nano-pure water
