Abstract
This dataset compiles a curated list of human seminal fluid proteins (SFPs) derived from two published proteomic studies of human seminal plasma. The first study contributes 879 high-confidence, non-redundant SFPs identified from three ejaculates of a single healthy donor (Pilch and Mann 2006), while the second contributes 1,923 non-redundant SFPs obtained from pooled individuals (Batruch et al. 2011). For each SFP-encoding gene, the dataset also includes the inferred evolutionary (phylogenetic) age of the gene (Chen et al. 2025) and its tau index, a measure of tissue-expression breadth (Palmer et al. 2021). This integrated resource is suitable for reuse in comparative genomic, evolutionary, and reproductive biology analyses, as well as studies exploring the tissue specificity of seminal proteome components. The dataset contains only summary proteomic and gene-level information derived from previously published studies, and does not include any directly identifying or individually identifiable information that could reasonably be used to identify the donors from whom the original samples were obtained.

Description of the data and file structure
The data in the file Dataset_S1.csv were obtained from 2 publications in which the authors characterized the proteomic content of human seminal plasma from semen samples. The first one includes a high-confidence subset of 879 non-redundant SFPs obtained from three ejaculates of a single healthy donor (Pilch and Mann 2006; DOI: 10.1186/gb-2006-7-5-r40). The second one relies on a set of non-redundant 1,923 human SFPs obtained from pooling different individuals (Batruch et al. 2011; DOI: 10.1021/pr100745u). With that information two sets of putative SFPs are provided, the common set and the reinforced set. The common set is just the intersection of the two non-redundant lists of human SFPs examined. The reinforced set includes the SFPs of the common set that passed the criteria of being identified by two peptides in both studies, plus the ≥10% sequence coverage threshold applied exclusively to the Batruch et al. (2011) data. The inferred evolutionary age of the SFP-encoding genes (Chen et al. 2025: DOI: 10.1101/2023.11.14.567139) and their tau index (Palmer et al. 2021; DOI: 10.18632/aging.202648) are also provided.
This dataset contains only proteomic and gene-level summary information derived from previously published studies. It does not contain any directly identifying or individually identifiable information, and cannot reasonably be used to identify the individual donors from whom the original semen samples were obtained.
Each of the two lists of SFP-encoding genes were examined for duplicate entries and then combined, resulting in one final expanded set.
