Description: This file contains the statistical output for the 72-hour MRI tracking experiment comparing CD4-labeled T-cell–treated mice with unlabeled T-cell controls. It summarizes changes in average T2 signal over time, including individual mouse trajectories, group-level comparisons, GLM/ANCOVA analyses, nonparametric Wilcoxon and Kruskal-Wallis tests, and time-specific t-tests. The file was used to evaluate whether CD4-labeled T cells produced a measurable T2 signal change relative to unlabeled controls during the early post-injection imaging window.
Page 1: Individual mouse trajectories for change from baseline T2 average across the 72-hour period. Red lines represent CD4-labeled T-cell mice and blue lines represent unlabeled controls. Pages 2–7: GLM/ANCOVA analysis of change-from-baseline T2_ave. The group effect is significant, with CD4-labeled T-cell mice showing a larger negative change than unlabeled controls. The model reports a group effect of approximately −2.34 with p < 0.0001, while the time effect is not significant when included as a covariate. Pages 8–10: Nonparametric comparisons at individual time points. Baseline/time 0 is not significant, 24 h shows a significant group difference by Wilcoxon/Kruskal-Wallis testing, and 72 h is not significant. Pages 11–18: Overall and time-specific t-test outputs, including group means, standard deviations, confidence intervals, variance tests, distribution plots, and Q-Q plots for checking distributional assumptions.

Description: Raw MRI data.
| Purpose / use | This workbook stores raw MRI T2 mapping measurements from CD4-labeled T cell tracking experiment. The dataset is intended for organizing T2 values, calculating average T2 values, and comparing the CD4 labeled T cell group with the unlabeled-control group over time. |
| Source data | The raw measurements are stored in Sheet1!A1:H145. Each mouse is represented by a block of repeated ROI or measurement rows. The mouse ID and mouse-level average values appear on the first row of each block; blank mouse-ID rows below belong to the same mouse block. |
| Timepoints | Prescan / baseline, 24 h after cell administration, and 72 h after cell administration. |
| Groups | CD4 labeled T cells vs. Unlabeled control. The workbook contains 8 mice per group, 16 mice total. |
| Unit note | T2 values are reported in milliseconds (ms). |
Code/software
Electron microscopy data: Selected areas were thin sectioned on a microtome at 60–80 nm, mounted on copper 300 mesh grids and then stained with uranyl acetate and lead citrate. Grids were examined using a FEI Talos F200X transmission electron microscope(Thermo Fisher Scientific, Waltham, MA, USA) and STEM and HAADF images and EDS spectra were acquired using inbuilt software and digital camera.
Raw MRI data: MRI images were acquired on the Bruker BioSpec 7T (Billerica, MA, USA). Axial images of mouse liver were acquired using a Turbo RARE sequence with parameters TR/TE = 2000/20 ms, Matrix size = 128 × 128, FOV = 36 × 24 mm2, Slice thickness = 1.2 mm, Averages = 2, Slice number = 16 and a T2map Multi Slice Multi Echo (MSME) sequence with TR = 2000 ms and 10 TEs equally spaced from 6.4 to 64 ms, Matrix size = 128 × 128, FOV = 36 × 24 mm2, Slice thickness = 1.2 mm, Averages = 3, Slice number = 16. T2 values were extracted using the system’s inbuilt Image Sequence Analysis (ISA T2vtr, Paravision 360, V3.0) software by defining three circular liver ROIs over 3 slices (9 measurements per mouse per time point). ROIs were drawn by a blinded, experienced radiologist across different liver Couinaud segments, avoiding large vessels such as the hepatic and portal venous branches. T2 maps were saved using the same software, and a custom color lookup table was created to aid visualization of the T2 values.
Functional flow cytometry data: All data were acquired on a BD LSR Fortessa X-20 flow cytometer (BD Biosciences, Franklin Lakes, NJ, USA) collecting ≥50,000 events per sample. Data were analyzed using FlowJo v10 (Tree Star, Ashland, OR, USA).
Statistical analysis: A one-way ANOVA model was fitted to analyze the change from baseline outcome by comparing the two groups at each time point. The results were consistent with those obtained from the two-sample t-test that was conducted. A non-parametric model, specifically the Wilcoxon test, was used to evaluate the fit of the above model and determine if the results will differ due to the small sample size. The findings from the non-parametric tests aligned with the earlier results. The standard 5% significance level was used for all hypothesis testing. All statistical analyses were performed using SAS software Version 9.4 (TS1M1, SAS Institute, Cary, NC, USA).
