Abstract
The data collected corresponds to experiments conducted using a standard hypoxia chamber with a cell culture set-up. The goal of the work was to create a mathematical model of oxygen transfer in the chamber. The collected data was used as an input to the model (temperature and inlet flow pattern data), used to inform model assumptions (evaporation data), and used to calibrate and validate the model (oxygen measurement and time delay data).

Description of the data and file structure
The data collected corresponds to experiments conducted using a standard hypoxia chamber with a cell culture set-up. The goal of the work was to create a mathematical model of oxygen transfer in the chamber. The collected data was used as an input to the model (temperature and inlet flow pattern data), used to inform model assumptions (evaporation data), and used to calibrate and validate the model (oxygen measurement and time delay data). The data is available in a single Excel file, with separate spreadsheets for each data set; there are a total of 10 spreadsheets.
Files and variables
File: Compiled_Data_PAPER_unformatted.xlsx
Description: The file Compiled_Data_Paper.xlsx contains data for temperature measurements (gas and liquid) for various positions in the chamber at different points in time, measurements of water evaporation from the cell culture wells, and oxygen measurements in the gas and liquid phases for both step and fluctuating changes in inlet oxygen. The oxygen measurement data was used for calibration and validation of the oxygen transfer model.
Variables
- Time (s)
- Temperature (degrees Celsius)
- Liquid Volume (mL)
- Gas phase oxygen (% of total gas in chamber)
- Liquid phase oxygen (μM)
- Inlet oxygen (fraction of total inlet)
Abbreviatons
- P1: Position 1 for oxygen measurement (defined in Paper)
- P2: Position 2 for oxygen measurement (defined in Paper)
- P3: Position 3 for oxygen measurement (defined in Paper)
Evaporation data: Sheet titled 'Water Bath Effect'
- Water Bath Effect: Data for volume remaining in cell culture wells after running gas chamber for 2 hours, with and without water in the water bath.
Temperature data: Sheets titled 'Temperature (No Water)','Temperature (With Water)', and 'Lid-Gas Temperature Difference'
- Temperature (No Water): Data for temperature measured at different locations in the gas chamber at various time points, in the absence of water in the cell culture wells and water bath. Measurement points include: the gas phase above each well, the gas phase directly above the water bath, and the gas phase above the chamber bottom.
- Temperature (With Water): Data for temperature measured at different locations in the gas chamber at various time points, with water in the cell culture wells. Measurement points include: the gas phase above each well, the gas phase directly above the water bath, the gas phase above the chamber bottom, and the water in each cell culture well.
- Lid-Gas Temperature Difference: Data for test runs with temperature measurements at random x-y locations in the chamber (measured at lid and directly below for each trial) over 2 hours. Also contains measurements (at lid and directly below) around each well at the 0.5-hour mark.
Time delay data: Sheet titled 'Measurement Delay'
- Time taken for gas and liquid probes to respond to step increase in inlet oxygen fraction to chamber.
Oxygen measurement data: Remaining sheets (collected data was used to make figures in the associated paper)
- Gas Phase Oxygen (Step Change): Oxygen measurements taken at 5-second intervals at 2 positions in the chamber, in response to a step increase in inlet oxygen fraction.
- Gas Phase Oxygen (IH): Oxygen measurements taken at 5-second intervals at 3 positions (and 1-second interval at 1 position) in the chamber, in response to fluctuating inlet oxygen fraction. Also contains flow data for the chamber inlet for each trial (inlet oxygen fraction recorded at approximately 1-second intervals).
- Liquid Phase (Step Change): Dissolved oxygen measurements taken at 5-second intervals at one cell culture well in the chamber, in response to a step decrease in inlet oxygen fraction. Also contains evaporation data for the volume of water remaining in the cell culture wells after 30 minutes of chamber run time.
- Liquid Phase (IH, No Cells): Dissolved oxygen measurements taken at 5-second intervals at one cell culture well in the chamber (without any seeded cells), in response to fluctuating inlet oxygen fraction. Also contains flow data for the chamber inlet for each trial (inlet oxygen fraction recorded at approximately 1-second intervals).
- Liquid Phase (IH, Cells): Dissolved oxygen measurements taken at 5-second intervals at one cell culture well in the chamber (with seeded endothelial cells), in response to fluctuating inlet oxygen fraction. Also contains flow data for the chamber inlet for each trial (inlet oxygen fraction recorded at approximately 1-second intervals).
