Abstract
Adult-born granule cells (abGCs) project to the CA2 region of the hippocampus, but no previous studies have assigned a behavioral function to this circuit. Here we show that abGC input to the CA2 is necessary for the retrieval of remote social memories. We also found that abGCs are necessary for differential CA2 network activity, including theta-gamma coupling and sharp wave-ripples, in response to novel versus familiar social stimuli.
README: Electrophysiology recordings (CA2):
The Electrophysiology.zip file contains raw data obtained from wireless hippocampal recordings in freely behaving mice. In one experiment, Nestin-cre and Nestin-cre:Gi mice were recorded at baseline and during exposure to a social stimulus. In another experiment, Nestin-cre and Nestin-cre:Gi mice were recorded at baseline and during exposure to a non-social stimulus. For both experiments, animals were recorded after vehicle or CNO administration. Drug administration was counterbalanced, as was the order of stimulus presentation. For the social stimulus experiment, recordings occurred when Gi-DREADD+ adult-born neurons were 4-6 weeks old and once again when the cohort of neurons was 10-12 weeks old. For the non-social stimulus experiment, recordings occurred when Gi-DREADD+ adult-born neurons were 4-6 weeks old only. Data filtering and normalization methods, as well as Python scripts used for analysis, are provided in the manuscript. Files were originally saved as .nex files before being converted to .mat files.

Code:
For remote social memory trials, all file names end with three letters (vtb, zuv, etc).
Letter #1: v = Nestin-cre, z = Nestin-cre:Gi\
Letter #2: t = mother stimulus, u = novel mother stimulus
Letter #3: v = male, b = female
For non-social object trials, all file names end with three letters (vob, zov, etc).
Letter #1: v = Nestin-cre, z = Nestin-cre:Gi
Letter #2: o = object trial
Letter #3: v = male, b = female
For each trial there are four separate files (ending in _1, _2, _3, and _4). These files contain recordings from the corresponding electrode implanted in CA2 (each animal had 4 electrode wires chronically implanted in CA2).
File names contain either "Veh" or "CNO" to indicate whether vehicle or Clozapine N-oxide were delivered, respectively.
Once opened, each matlab file has three columns.

Histology
The Histology.zip file contain images of the dorsal hippocampus used for analysis in the manuscript. Sections were stained with RGS14, PCP4, 3R-Tau, and Znt3 antibodies. Files can be opened using Fiji (ImageJ).
Histology.zip contains a folder called "Histology" with the following files:
CD1 and GFAP-TK Recovery tissue - Hippocampus images from CD1 and GFAP-TK animals six weeks after VGCV cessation.
GFAP-TK VGCV+ tissue - Hippocampus images from GFAP-TK mice after VGCV administration.
Code.xlsx - contains two tabs: 1) CD1 + GFAP-TK Recovery and 2) GFAP-TK VGCV. Tabs decode sex, strain, and condition for animals used for histological analyses.
